体外研究

L-660,711(MK-571)在豚鼠肺中抑制[3H]LTD4的结合，Ki值为0.22±0.15 nM (n=35)，在人类肺部抑制[3H]LTD4的结合，Ki值为2.1±1.8 nM (n=29)。L660,711对[3H]LTC4的结合活性较低或没有活性，在豚鼠肺部IC50为23±11 μM (n=16)、在人类肺部中IC50为32 μM (n=1)。
体内研究

健康年轻人对MK-571耐受良好。在口服后，MK-571被迅速吸收。在给药后1.1-1.5小时后血药浓度达到峰值。

方法

将细胞以1×103/孔的密度接种于96孔板，在37℃、5% CO2的细胞培养箱中培养72小时，让MRP1的信使RNA受到抑制。然后用对照培养基或化疗药物对细胞进行处理，处理72小时。之后，将溶解于PBS中的MTT加入到每孔，孵育4小时，移去MTT溶液，加入DMSO。10分钟后，测定570 nm处的吸光值。在进一步化疗药物处理前或检测前7小时，将小分子抑制剂MK571（25 μM）和Reversan (15µM)加入其中。
