TrkB受体,又称TrkB酪氨酸激酶或BDNF/NT-3生长因子受体或神经营养性酪氨酸激酶受体2型(NTRK2),是一种具有胞内酪氨酸激酶活性的单次跨膜催化受体。TrkB/NTRK2属于神经营养性酪氨酸受体激酶(NTRK)家族,包含两个Ig样C2型结构域、两个LRR(富含亮氨酸)重复序列、一个LRRCT结构域、一个LRRNT结构域及一个蛋白激酶结构域。TrkB/NTRK2酪氨酸激酶与Ras、Cdc42/Rac/RhoG、MAPK、PI3-K和PLCγ信号通路相偶联。TrkB/NTRK对脑源性神经营养因子(BDNF)具有高亲和力,参与神经元可塑性、长时程增强及中枢神经系统神经元的凋亡过程。作为膜结合受体,TrkB/NTRK与神经营养因子结合后发生自磷酸化,并磷酸化MAPK通路成员。NTRK2/TrkB通过调控神经元存活、增殖、迁移、分化及突触形成与可塑性,参与中枢与周围神经系统的发育和成熟。TrkB/NTRK的突变与肥胖及情绪障碍相关。
