背景
VEGF-165 是血管内皮生长因子A(VEGF-A)最主要的剪接变体。它由多种细胞产生,包括血管内皮细胞、巨噬细胞和T细胞。VEGF-165 参与血管生成、调控血管内皮细胞的存活、生长、迁移以及血管通透性。VEGF基因的表达受缺氧、炎症细胞因子和癌基因诱导。VEGF-165可与硫酸乙酰肝素结合,从而滞留于细胞表面和细胞外基质中。它还与受体酪氨酸激酶VEGFR1和VEGFR2结合。独特的是,VEGF-165是唯一能够与辅助受体神经纤毛蛋白-1(NRP-1)和神经纤毛蛋白-2(NRP-2)结合的剪接变体,这些辅助受体起到增强VEGFR2信号通路的作用。VEGF-165与VEGFR1和VEGFR2的结合,可激活包括PI3K/Akt、p38 MAPK、黏着斑激酶和桩蛋白在内的多条信号通路。VEGF在多种癌症的肿瘤血管生成过程中扮演关键角色。
