| 背景 | Nedd4-2 is a ubiquitin protein ligase of the Nedd4 family which is known to bind and regulate a number of membrane proteins to aid in their internalization and turnover. Several of the NEDD4-2 substrates include ion channels, such as the epithelial and voltage-gated sodium channels. NEDD4-2 plays critial roles in regulating membrane proteins and is essential for the maintenance of cellular homeostasis. Nedd4-2 is an E3 ubiquitin-protein ligase that reduces expression of the epithelial Na+ channel ENaC at the cell surface. Nedd4-2 catalyzes ubiquitination of α-, β-, and γENaC. Its overexpression increases ubiquitination, whereas Nedd4-2 silencing decreases ubiquitination. |
