| 体外研究活性 | ZSTK474在1 μM浓度下有效将PI3K活性降低为对照组水平的4.7%，而LY2194002仅将其活性降低为对照组的44.6%。ZSTK474抑制重组p110β，-γ，和 -δ活性，IC50 分别为17 nM，53 nM，和6 nM。ZSTK474对一组39个人类癌细胞系表现出有效的抗增殖活性，平均GI50为0.32 μM，比平均GI50 分别为7.4 μM和10 μM的LY294002与wortmannin更有效。1 μM ZSTK474治疗阻断小鼠胚胎成纤维细胞(MEFs)中血小板源生长因子诱导的胞膜边缘波动和PIP3产生。10 μM ZSTK474诱导OVCAR3细胞凋亡，并诱导细胞周期完全的G1期阻滞，但是不引起A549细胞凋亡。0.5 μM ZSTK474治疗显著减少磷酸化的Akt 和GSK-3β水平，以及细胞周期蛋白D1蛋白质的表达。ZSTK474也会剂量依赖性抑制其他参与细胞增殖调节的下游信号组分，包括FKHRL1，FKHR，TSC-2，mTOR，和p70S6K的磷酸化。[1] ZSTK474在0.1 μM下不抑制mTOR，甚至在100 μM浓度下，ZSTK474仅抑制少于40%的mTOR活性。[2] ZSTK474阻断人脐静脉内皮细胞(HUVECs)中VEGF诱导的细胞迁移和管腔形成，并且抑制RXF-631L细胞中HIF-1α表达与VEGF分泌，表现出有效的体外抗血管生成活性。[3] ZSTK474治疗抑制伴刀豆球蛋白A激活的T细胞中IFNγ 和IL-17的产生，并通过成纤维细胞样滑膜细胞(FLS)抑制增殖和PGE(2)产生。[6] |
